Tram route 75 is a public transport service in Melbourne, Victoria, Australia. It is operated by Yarra Trams and runs from the corner of Spencer Street and LaTrobe Street in the city centre to Vermont South in the eastern suburbs.

Unike most tram routes in Melbourne, it is not entirely within Metcard ticketing Zone 1. The portion of the route between Camberwell Junction (corner of Riversdale, Camberwell and Bourke Roads) and Warrigal Road is a Zone 1 and Zone 2 overlap, and everything East of Warrigal Road is Zone 2 only. Between Springvale Road and Vermont South is a Zone 2 and 3 overlap. A connecting bus, the Knox Transit Link (operated by Ventura Bus Lines using buses from [Route 732]), runs to Knox City meeting every tram and also runs within Zone 2 and Zone 3. It is planned to further extend the tram line to Knox City Shopping Centre, replacing the bus.

The route runs along Flinders Street in the city, Bridge Road in Richmond, Riversdale Road through Hawthorn, Camberwell and Toorak Roads through Camberwell and then along Burwood Highway, past Deakin University, to the terminus in Vermont South. Prior to July 2005 the route terminated at Burwood East. The final three kilometres of track and a new Vermont South Terminus at the shopping centre were added then.
