In biology, a pure culture is a population of identical cells originating from a single cell.

Pure cultures are obtained by working in aseptic conditions by growing isolated colonies of microorganisms, usually on a petri dish. The petri dish contains an appropriate sterile medium for the microorganism of interest. The initial sample is diluted progressively using various techniques to eventually have some areas of the petri dish with a single cell. The petri dish is incubated long enough in appropriate environmental conditions until visible isolated colonies appear.
