His best known book, The Hot Zone (1994), is a nonfiction bio-thriller about the Ebola virus. He came to know the ebola virus through many contacts including C.J Peters and Nancy Jaax. His facination began during a visit to Africa and people were dropping like flies.

Richard Preston's fictional novel "Cobra Event", dealing with the terroristic release of a fictional virus composed of various qualities of true diseases in New York, alarmed even former President Clinton.
