Sub-cloning

Sub-cloning is the method by which two distinct gene(s) or gene fragments are fused together by means of restriction sites. Typically, a genetic insert from one vector is inserted into another vector. This technique is related to but distinct from "cloning". The procedure involves releasing and purifying the target insert from a parent vector, ligating this insert into a destination vector and transforming this ligation reaction into competent bacterial cells. Finally, the transformed cells are screened for the target vector to confirm successful transfer.
